What Selank nasal spray is
Tuftsin is a four-residue fragment, Thr-Lys-Pro-Arg, clipped from immunoglobulin G. Selank keeps that core and adds a Pro-Gly-Pro tail. Researchers at Russia's Institute of Molecular Genetics used the same tail on Semax, a different parent peptide, to slow carboxypeptidase attack.[2] The molecule in this bottle is a free carboxylic acid at the C-terminus.
It is the same heptapeptide as the lyophilized Selank vials. The format is not. This SKU is a clear nasal solution, one 10 mg bottle, $79, no 5-pack, no 10-pack, no second strength. Appearance on the specs table should read as a spray, not a lyophilized puck.
Some catalogs list N-Acetyl Selank Amidate next to Selank, often in a 30 mg vial. That analog adds an acetyl group at the N-terminus and an amide in place of the C-terminal acid. Why so much of the Selank literature is a spray
The registered Russian product is an intranasal solution. Once a peptide has a nasal drug product, later papers tend to keep using that route. The 2008 GAD comparison sits in that pharmaceutical context. The 2008 hippocampus paper dosed rats in the nose on purpose.[5][6]
Labs pick the olfactory and trigeminal routes when they want material near CNS tissue without a systemic injection. That is a study-design choice. It is not a pharmacokinetic map for Tetrava's 10 mg research spray, and it does not make a spray bottle the right tool for an in-vitro enzyme plate.
What does Selank nasal spray do
The papers describe enzyme kinetics, gene panels, a rat BDNF assay, and one Russian clinical comparison. They do not describe an outcome from this research bottle used on a person.
The 2001 mechanism paper measured how fast leu-enkephalin, an endogenous opioid peptide, is cleared from plasma. Patients with generalized anxiety disorder had a shorter enkephalin half-life and lower enkephalinase-inhibiting activity than healthy controls. Selank added in vitro blocked the peptidases that cut leu-enkephalin, with an IC50 around 15 micromolar, and beat the reference inhibitors bacitracin and puromycin at that job.[3]
The proposed chain is simple enough: slower enzymatic breakdown leaves enkephalin in circulation longer. That is an in-vitro enzyme assay plus a correlational blood finding. It is not a receptor-binding study showing Selank sitting on an opioid receptor.
GABAergic gene expression, and a cell-culture check
A 2016 rat study compared Selank with GABA itself. The authors ran a real-time PCR panel of 84 genes tied to neuroreception and neurotransmission, including GABA receptor subunits, transporters, ion channels, and dopamine and serotonin receptors, in frontal cortex at 1 hour and 3 hours after 300 micrograms per kilogram of Selank or GABA.[4] Forty-five genes shifted at one hour. Twenty-two shifted at three hours. Expression changes after Selank tracked the changes after GABA at the one-hour mark.
That paper does not, in the English abstract, specify a nasal route. Treat it as a gene-expression argument for a GABAergic component, not as a spray-device paper.
A 2017 follow-up put Selank on human neuroblastoma IMR-32 cells and measured the same class of GABAergic transcripts. Selank alone did not change those mRNA levels. Combined with GABA, it almost wiped out the expression shifts GABA produced on its own. Combined with olanzapine, more genes moved than with olanzapine alone.[7] The peptide did not look like a direct transcriptional switch in that cell line. The authors still argued it can change how GABA interacts with GABA-A receptors. Two models, two answers. A lab that needs one of them should pick the model, not a forum summary of both.
Selank nasal spray for anxiety: the 2008 comparison
The trial behind most "Selank nasal spray for anxiety" copy enrolled 62 patients with generalized anxiety disorder or neurasthenia. Thirty got Selank. Thirty-two got medazepam, a benzodiazepine. Outcomes used Hamilton, Zung, and Clinical Global Impression scales, plus serum enkephalin activity.[6]
Anxiolytic scores were similar. The Selank arm also showed antiasthenic and psychostimulant effects that the English abstract does not report for medazepam. Patients arrived with a shortened leu-enkephalin half-life that tracked disease duration, anxiety and asthenia severity, and autonomic symptoms. That half-life rose during Selank treatment, more clearly in the GAD subgroup, and the correlation with anxiety scores got stronger.
PubMed indexes the paper as a randomized controlled trial and tags both intranasal and oral administration. The abstract does not publish a placebo arm. One country, one language of record, no FDA review. Russia's 2009 0.15% nasal registration sits on this evidence. The study reported that the intranasal route changed hippocampal BDNF in vivo.

Tuftsin's first research home was immunology, phagocyte activity and cytokine signaling, not anxiety. Some of that interest carried into Selank. A 2021 social-stress study in rats measured serum cytokines after 20 days of brief daily male confrontations. Stressed animals had higher IL-1beta, IL-6, and TGF-beta1. Selank given intraperitoneally at 100 micrograms per kilogram per day brought those readings back toward control values, with IL-4 restored and TNF-alpha suppressed.[8]
That paper used an intraperitoneal dose, not a nasal spray. It is a rodent cytokine panel, thinner than the enkephalinase or 2016 gene-expression work, and it is not a human immunology trial.
The FDA has never approved Selank in any form. In July 2026 the Pharmacy Compounding Advisory Committee reviewed seven peptide bulk substances nominated for the Section 503A Bulks List.
