Semax Nasal Spray 10mg Peptide Research

What Semax nasal spray is

Adrenocorticotropic hormone has a short internal stretch, residues 4 through 7: Met-Glu-His-Phe. Researchers at Russia's Institute of Molecular Genetics trimmed that stretch from ACTH(4-10) and stuck a Pro-Gly-Pro tail on the end. The tail slows carboxypeptidase attack. The same tail later went onto Selank, which is built from a different parent peptide.[2]

Seven residues. A free carboxylic acid at the C-terminus. That is the molecule in this bottle.

It is the same heptapeptide as the lyophilized Semax vials. The container is different. This SKU is a clear nasal solution, one 10 mg bottle, $79. No 5-pack. No 10-pack. No second strength. The specs table should read as a spray, not a white lyophilized puck.

N-Acetyl Semax and N-Acetyl Semax Amidate add an acetyl group at the N-terminus, an amide in place of the C-terminal acid, or both. Other vendors often sell those as a 30 mg analog. Why so much of the Semax literature is a spray

The registered Russian product is an intranasal solution. Once a peptide has a nasal drug product, later papers tend to keep using that route.

Dolotov's 2006 hippocampal BDNF paper used a single intranasal dose. Romanova's cortical-infarct study dosed rats in the nose for six days. Polunin's optic-nerve trial compared intranasal drops with endonasal electrophoresis.[3][4][11] A second 2006 Dolotov paper, in Journal of Neurochemistry, also used the nose: 50 and 250 micrograms per kilogram, then BDNF protein in basal forebrain at three hours.[13]

Labs pick the olfactory and trigeminal routes when they want material near CNS tissue without a systemic injection. That choice belongs to the protocol. It does not turn Tetrava's 10 mg research spray into a pharmacokinetic map, and it does not make a spray bottle the right tool for an in-vitro enzyme plate.

What does Semax nasal spray do

"Semax nasal spray benefits" usually wants a human outcome. The papers have endpoints: BDNF protein, trkB phosphorylation, infarct volume, conditioned avoidance, optic-nerve scores. Those are study readouts. They are not a result from this research bottle used on a person.

The mechanism thread that gets cited most is gene expression, not a single named receptor. Shadrina 2010 compared BDNF and NGF mRNA timing across rat hippocampus, frontal cortex, and retina. The transcripts moved on different clocks. Hippocampus responded first.[5] BDNF is a neuron-survival and synaptic-plasticity readout in those models. NGF is the cholinergic-neuron one.

The same year as the hippocampal fold-change paper, Dolotov's group asked a more basic question: does the basal forebrain even have a binding site? Tritium-labelled Semax bound rat basal-forebrain membranes in a time-dependent, specific, reversible way. Binding needed calcium ions. They reported a KD of 2.4 nM (SEM 1.0) and a Bmax of 33.5 fmol/mg protein (SEM 7.9).[13] Then they put the peptide in the nose at 50 and 250 micrograms per kilogram. BDNF protein was up at three hours in basal forebrain. Cerebellum did not move. The authors had already seen Semax raise BDNF synthesis in basal-forebrain astrocytes in culture. This experiment asked whether the intact rat would show it after an intranasal dose.

N-Acetyl Semax nasal spray is a different catalog item

Search results for N-Acetyl Semax nasal spray, Semax amidate, and Adamax sit next to unmodified Semax because the names share a stem. Acetylation and C-terminal amidation change how peptidases clip the chain. None of the papers cited on this page is a head-to-head of N-Acetyl Semax against this unmodified spray.

If the protocol specifies the acetylated analog, use Adamax and that page's identity data. If it specifies native Semax as a finished spray, this is the listing. A 30 mg amidate vial from another vendor is not this 10 mg bottle.

Does Semax nasal spray work: the 2006 hippocampal BDNF paper

Dolotov, Karpenko, Inozemtseva and colleagues gave rats a single 50 micrograms per kilogram intranasal dose of Semax and then measured the hippocampal BDNF/trkB system. BDNF protein rose to a maximum of 1.4-fold. TrkB tyrosine phosphorylation rose 1.6-fold. Exon III BDNF mRNA rose 3-fold. TrkB mRNA rose 2-fold. The same animals showed more conditioned avoidance reactions than controls.[3]

Use this paper when the assay question is a single intranasal dose and hippocampal protein plus phosphorylation. Use Shadrina 2010 when the question is BDNF and NGF mRNA on different clocks in hippocampus, cortex, and retina.[5] Use the Journal of Neurochemistry paper when the question is a basal-forebrain binding site and a three-hour BDNF protein rise that did not appear in cerebellum.[13]

Those are rat protocols with fixed timepoints. They are not a timer for when a person would feel something.

Cerebral ischemia and optic-nerve papers that used a nasal route

Romanova 2006 used photochemically induced prefrontal infarction in rats. Six days of intranasal Semax reduced infarct volume and improved passive-avoidance performance against untreated controls.[4]

Two later papers went into the same ischemia-reperfusion biology without turning this spray into a drug. Filippenkov 2020 counted 394 differentially expressed genes after transient middle-cerebral-artery occlusion. Inflammation-related transcripts went down relative to saline. Neurotransmission-related transcripts went up.[6] Sudarkina 2021 measured protein instead of RNA. Active CREB went up. MMP-9, c-Fos, and active JNK went down.[7] Transcriptome, then proteome. Rodent. Not a human dose-response for this bottle.

A 2000 Vestnik Oftalmologii trial tested Semax in vascular, toxic-allergic, and inflammatory optic-nerve disease, including partial optic-nerve atrophy. Three groups: intranasal drops, endonasal electrophoresis, untreated control. Both treated arms were tied to better visual acuity, a wider visual field, better electrical conductivity through the optic nerve, and improved color vision relative to the control group.[11] One Russian trial. No independent Western replication.

Gusev 2018 is the other human line most often cited, a series on patients treated at different stages of ischemic stroke. PubMed indexes it as a clinical trial. Same single-health-system limit.[12]

Pentapeptide fragments were the main byproduct. Glial and neuronal cells produced slightly different fragment patterns from the same starting peptide.[10]

That map is about how the chain gets cut in tissue culture. It is not "how long until it works." Dolotov's hippocampal paper used a single rat dose and a BDNF readout. The basal-forebrain paper used a three-hour protein timepoint. Those clocks belong to the experiments.
